TMEM243 is a mitochondrial protein predicted to play a role in drug resistance in cancer (Duan et al., 2004).
▼ Cloning and Expression
Using microarray analysis to identify genes upregulated in taxol- and doxorubicin-resistant human cancer cell lines, Duan et al. (2004) identified TMEM243, which they called MMTRAG. The deduced 118-amino acid protein has a calculated molecular mass of 13.4 kD. RNA dot blot analysis detected wide expression of MMTRAG in normal and neoplastic tissues. In normal tissues, MMTRAG expression was relatively high in lung, kidney, and thyroid gland. Fluorescence-tagged MMTRAG localized to mitochondria in transfected U2OS cells.

By Northern blot analysis, Duan et al. (2004) found that 7 of 8 taxol- or doxorubicin-resistant human cancer cell lines overexpressed MMTRAG. Expression profiling revealed elevated MMTRAG expression in a high number of taxol- and doxorubicin-treated cancer cell lines.
