▼ Cloning and Expression
By searching EST databases for sequences similar to OATP (SLCO1A2; 602883), followed by RT-PCR of adult brain RNA and screening a kidney cDNA library, Tamai et al. (2000) cloned SLCO3A1, which they designated OATPD. The deduced 710-amino acid protein has 12 potential transmembrane regions and several phosphorylation and N-glycosylation sites. RT-PCR detected OATPD expression in all adult and fetal tissues examined except adult skeletal muscle.

By PCR of a brain cDNA library, Huber et al. (2007) isolated 2 SLCO3A1 splice variants that the designated OATP3A1-V1 and -V2. OATP3A1-V1 and -V2 differ in the splicing of exon 10 and encode deduced proteins of 710 and 692 amino acids, respectively, that are identical over the first 666 amino acids. Real-time PCR detected widespread expression of OATP3A1-V1 and generally lower expression of OATP3A1-V2, with predominant OATP3A1-V2 expression in testis and brain. Immunohistochemical analysis detected OATP3A1-V1 in testicular germ cells, the basolateral plasma membrane of choroid plexus epithelial cells, and neuroglial cells of human frontal cortex gray matter. OATP3A1-V2 immunolocalized to Sertoli cells in testis, apical and/or subapical membranes in choroid plexus epithelial cells, and in all cell bodies and axons of neurons of frontal cortex gray and white matter.

Tamai et al. (2000) overexpressed OATPD in HEK293 cells and found that it mediated transport of estrone-3-sulfate and, more weakly, prostaglandin E2.

Using cRNA-injected Xenopus oocytes and stably transfected Chinese hamster ovary cells, Huber et al. (2007) found that both OATP3A1-V1 and -V2 transported prostaglandins E1 and E2, thyroxine, a cyclic endothelin receptor (see ENDRA; 131243) peptide antagonist, and vasopressin (192340).

Huber et al. (2007) determined that the SLCO3A1 gene contains 11 exons and spans about 219 kb.

▼ Mapping
By radiation hybrid analysis, Tamai et al. (2000) mapped the SLCO3A1 gene to chromosome 15, between markers D15S202 and D15S157. Huber et al. (2007) stated that the SLCO3A1 gene maps to chromosome 15q26.
