▼ Description
GRID1 is subunit of glutamate receptor channels. These channels mediate most of the fast excitatory synaptic transmission in the central nervous system and play key roles in synaptic plasticity (Yamazaki et al., 1992).

▼ Cloning and Expression
Yamazaki et al. (1992) cloned mouse Grid1. The deduced 994-amino acid protein has an N-terminal signal peptide and 4 putative C-terminal transmembrane segments. The putative cytoplasmic domain between transmembrane segments 3 and 4 contains potential phosphorylation sites for calcium/calmodulin-dependent protein kinase II (see CAMK2A; 114078) and protein kinase C (see PRKCA; 176960).

By sequencing clones obtained from a size-fractionated fetal brain cDNA library, Nagase et al. (1999) cloned GRID1, which they designated KIAA1220. The deduced 791-amino acid protein shares 98% identity with mouse Grid1. RT-PCR ELISA detected high expression in adult brain, intermediate expression in fetal brain, low expression in testis and ovary, and no expression in all other peripheral tissues examined. All specific brain regions examined showed intermediate GRID1 expression.

By radiation hybrid analysis, Nagase et al. (1999) mapped the GRID1 gene to chromosome 10.

Treadaway and Zuo (1998) mapped the mouse Grid1 gene to a region of chromosome 14 that shares homology of synteny with human chromosome 10q11-q23.
