▼ Description

The poly(rC)-binding proteins are a subfamily of KH-domain-containing RNA-binding proteins that bind C-rich pyrimidine tracts and play pivotal roles in a broad spectrum of posttranscriptional events (Chkheidze and Liebhaber, 2003).

▼ Cloning and Expression

Based on its sequence similarity with mouse and human PCBP2 (601210), Makeyev and Liebhaber (2000) cloned mouse Pcbp3, and using this sequence as query to search an EST database, they identified human PCBP3. Both mouse and human PCBP3 encode 339-amino acid proteins with calculated molecular masses of 36 kD. With the exception of 5 amino acid differences, the mouse and human proteins are identical. PCBP3 contains 2 KH (HNRNPK (600712)-homology) domains in the N-terminal half, and 1 near the C-terminus. Quantitative RT-PCR detected expression in all mouse tissues examined, with highest levels in testis and fat, and the lowest level in heart. Chkheidze and Liebhaber (2003) found that epitope-tagged PCBP3 localized to the cytosol of transfected HeLa cells.

Makeyev and Liebhaber (2000) determined that the PCBP3 gene contains at least 14 exons and spans more than 93 kb.

▼ Mapping

By genomic sequence analysis, Makeyev and Liebhaber (2000) mapped the PCBP3 gene to chromosome 21q22.3. By FISH, they mapped the mouse Pcbp3 gene to a syntenic region of mouse chromosome 10B5.
